ABSTRACT

Myrtus communis L., commonly known as true myrtle, is a medicinal plant native to the Mediterranean area. Since ancient times, the inhabitant s of this area have been using it for its cultural and medicinal properties. Because of the vast diversity of biomolecules in its aerial parts, it exhibits several biological properties, including antioxidant, antimicrobial, and anticancer properties. This review retrospect the research on the source, biological activities with empirical evidence, chemical composition, applications, and cellular targets of extracts and essential oils obtained from M. communis leaves, which provides a perspective for further studies on the applications and formulations of extract and EO of M. communis leaves. The efficacy of constituents' individually, in association with other bioactive constituents, or in combination with available commercial drugs would provide insights in to the development of these bio - actives as future drugs and their evolving future potential applications in the pharmaceutical, food, and aroma industries.


Myrtus communis L., comúnmente conocido como arrayán verdadero, es una planta medicinal originaria de la zona mediterránea. Desde la antigüedad, los habitantes de esta zona lo utilizan por sus propiedades culturales y medicinales. Debido a la gran div ersidad de biomoléculas en sus partes aéreas, exhibe varias propiedades biológicas, incluidas propiedades antioxidantes, antimicrobianas y anticancerígenas. Esta revisión retrospectiva de la investigación sobre la fuente, las actividades biológicas con evi dencia empírica, la composición química, las aplicaciones y los objetivos celulares de los extractos y aceites esenciales obtenidos de las hojas de M. communis , lo que brinda una perspectiva para futuros estudios sobre las aplicaciones y formulaciones de l os extractos y EO de M. communis . La eficacia de los componentes individualmente, en asociación con otros componentes bioactivos o en combinación con medicamentos comerciales disponibles proporcionaría información sobre el desarrollo de estos bioactivos co mo medicamentos futuros y sus futuras aplicaciones potenciales en las industrias farmacéutica, alimentaria y aromática

ABSTRACT

Patchoulol is an important sesquiterpenoid in the volatile oil of Pogostemon cablin, and is also considered to be the main contributing component to the pharmacological efficacy and fragrance of P. cablin oil, which has antibacterial, antitumor, antioxidant, and other biological activities. Currently, patchoulol and its essential oil blends are in high demand worldwide, but the traditional plant extraction method has many problems such as wasting land and polluting the environment. Therefore, there is an urgent need for a new method to produce patchoulol efficiently and at low cost. To broaden the production method of patchouli and achieve the heterologous production of patchoulol in Saccharomyces cerevisiae, the patchoulol synthase(PS) gene from P. cablin was codon optimized and placed under the inducible strong promoter GAL1 to transfer into the yeast platform strain YTT-T5, thereby obtaining strain PS00 with the production of(4.0±0.3) mg·L~(-1) patchoulol. To improve the conversion rate, this study used protein fusion method to fuse SmFPS gene from Salvia miltiorrhiza with PS gene, leading to increase the yield of patchoulol to(100.9±7.4) mg·L~(-1) by 25-folds. By further optimizing the copy number of the fusion gene, the yield of patchoulol was increased by 90% to(191.1±32.7) mg·L~(-1). By optimizing the fermentation process, the strain was able to achieve a patchouli yield of 2.1 g·L~(-1) in a high-density fermentation system, which was the highest yield so far. This study provides an important basis for the green production of patchoulol.

ABSTRACT

Basil (Ocimum basilicumL.) is a medicinal species used in several areas, such as food, medicines and cosmetics, and the understanding of its physiological behavior under environmental conditions is of paramount importance for the improvement of cultivation methods. The objective of this study was to evaluate the influence of different water availability under physiological, biochemical and metabolic characteristics, in three distinct genotypes: 'Alfavaca basilicão', 'Gennaro de menta' and 'Grecco à palla', during two different phenological stages (vegetative and reproductive). It was found that the water deficit promotes physiological changes to tolerate water stress, and the studied genotypes have different routes to achieve this physiological tolerance, which culminates in a distinct accumulation of metabolites in plants, and can be considered interesting if the final product is the production of essential oils.


La albahaca (Ocimum basilicum L.) es una planta medicinal utilizada en varias áreas: alimenticia, medicinal e industria cosmética; es de suma importancia el entendimiento de su comportamiento fisiológico bajo diferentes condiciones ambientales con el fin de mejorar los procesos del cultivo. El objetivo de este estudio fue evaluar la influencia de diferentes disponibilidades hídricas en las características fisiológicas, bioquímicas y metabólicas en tres genotipos de albahaca: "Alfavaca basilicão", "Gennaro de menta" y "Grecco à palla" durante dos etapas fenológicas (vegetativa y reproductiva). Fue encontrado que el déficit hídrico promueve cambios fisiológicos con el fin de tolerar el estrés hídrico. Los genotipos estudiados presentaron diferentes rutas para alcanzar esta tolerancia fisiológica, la cual culmina con distintas acumulaciones de metabolitos en las plantas, y puede ser considerado interesante si el producto final es la producción de aceites esenciales.

Introducción: Las propiedades antibacterianas de Plantago major frente a microorganismos orales no se ha estudiado ampliamente. Objetivo: Identificar los metabolitos secundarios presentes en el extracto etanólico de Plantago major y determinar su actividad antibacteriana frente a Streptococcus mutans. Métodos: Se prepararon concentraciones del 25 por ciento, 50 por ciento, 75 por ciento y 100 por ciento de extracto etanólico (EE) de Plantago major. Se cargaron seis discos de papel con 10 µL, 15 µL, 20 µL y 25 µL de cada concentración, haciendo un total de 96 discos. Los discos fueron colocados en placas Petri con agar cerebro-corazón inoculadas con Streptococcus mutans, se empleó clorhexidina al 0,12 por ciento como control positivo. Se calcularon seis repeticiones para cada concentración. Las placas fueron incubadas a 37 °C por 48 horas. La medición de los halos de inhibición se realizó con un compás digital. El análisis estadístico se realizó mediante la prueba ANOVA de un factor seguido por la prueba post hoc de Tukey. Resultados: El análisis químico del extracto etanólico de Plantago major identificó la presencia de terpenos, diterpenos, saponinas, terpenoidales y aceites esenciales. Las concentraciones del 25 por ciento y 50 por ciento no mostraron efecto antibacteriano, los volúmenes mayores de 20 µL de la concentración del 75 por ciento y todas los del 100 por ciento fueron efectivos para inhibir el crecimiento de Streptococcus mutans con halos de inhibición de 8,36 mm a 14,64 mm. La clorhexidina al 0,12 por ciento inhibió el crecimiento de Streptococcus mutans con halos de inhibición de 17,77 mm en promedio, presentando diferencias significativas con todas las concentraciones del extracto etanólico de Plantago major (P < 0,05). Conclusiones: El extracto etanólico de Plantago major presentó derivados de los terpenos y saponinas, y mostró actividad antibacteriana frente a Streptococcus mutans en volúmenes y concentraciones mayores a 20 µL/75 por ciento(AU)


Introduction: The antibacterial properties of Plantago major against oral microorganisms have not been widely studied. Objective: Identify the secondary metabolites present in an ethanolic extract of Plantago major and determine their antibacterial activity against Streptococcus mutans. Methods: The Plantago major ethanolic extract (EE) was prepared at concentrations of 25 percent, 50 percent, 75 percent and 100 percent. Six paper discs were loaded with 10 µl, 15 µl, 20 µl and 25 µl of each concentration, for a total 96 discs, which were then placed on Petri plates with brain heart agar inoculated with Streptococcus mutans. The positive control was 0.12 percent chlorhexidine. Six replicates were estimated for each concentration. The plates were incubated at 37ºC for 48 hours. Inhibition haloes were measured with a digital caliper. Statistical analysis was based on one-factor ANOVA testing followed by Tukey's post hoc test. Results: Chemical analysis of the Plantago major ethanolic extract identified the presence of terpenes, diterpenes, saponins, terpenoids and essential oils. The 25 percent and 50 percent concentrations did not display an antibacterial effect, whereas volumes above 20 µl of the 75 percent concentration and all 100 percent volumes were effective to inhibit Streptococcus mutans growth with inhibition haloes of 8.36 mm to 14.64 mm. 0.12 percent chlorhexidine inhibited Streptococcus mutans with inhibition haloes of 17.77 mm on average, presenting significant differences with all the concentrations of the Plantago major ethanolic extract (p < 0.05). Conclusions: The Plantago major ethanolic extract was found to contain terpene and saponin derivatives, and displayed antibacterial activity against Streptococcus mutans at volumes and concentrations above 20 µl / 75 percent(AU)

ABSTRACT

The objective was to evaluate the production and composition of lavender essential oil, simultaneously at open field and greenhouse, in spring-summer season, under different types of fertilization. Each cultivation system was considered an experiment (field and greenhouse), and in both, the experimental design used was randomized blocks, with five treatments, being fertilization with nutrient source -100% mineral (100%M), 100% organic (100%O), 100% organomineral (100%OM), 50% organomineral (50%OM), and without fertilization (control), and three repetitions. Better performance and productivity of Lavandula dentatais related to respectively 100% OM and 50% OM fertilization in the field and greenhouse. The essential oil content does not differ between cultivation systems and kind of fertilizers, and the majorities compounds were 1.8-cineol, fenchone and camphor.


El objetivo fue evaluar la producción y composición del aceite esencial de lavanda a campo abierto e invernadero simultáneamente, en la temporada primavera-verano, bajo diferentes tipos de fertilización. Cada sistema de cultivo fue considerado un experimento (campo e invernadero), y en ambos, el diseño experimental utilizado fue bloques al azar, con cinco tratamientos, siendo la fertilización con fuente de nutrientes 100% mineral (100% M), 100% orgánico (100% O), 100% organomineral (100% MO), 50% organomineral (50% MO), y sin fertilización (control), y tres repeticiones. Un mejor rendimiento y productividad de Lavandula dentata está relacionado con una fertilización de 100%OM y 50% OM en el campo y en invernadero, respectivamente. El contenido de aceite esencial no difiere entre sistemas de cultivo y tipo de fertilizante, y los compuestos mayoritarios fueron 1.8-cineol, fenchone y alcanfor.

ABSTRACT

Volatiles compounds are involved in defensive induction against insects, playing an important role in insect-plant interaction being induced by response to mechanical damage. However, they could decrease according to the domestication degree in cultivated plants. Currently, it has been established that secondary metabolites are reduced due to the domestication process in murtilla. Hence, the follow question emerges: Are volatile organic compounds induced by mechanical damage reduced in cultivated murtilla plants in relation to wild plants? Two cultivated ecotypes and their respective wild counterparts were sampled. Volatiles compounds were obtained using Porapak-Q columns and analyzed by gas chromatography. Results showed that compounds as 2-hexanone, α-pinene, 2-thujene, 3-thujene and 1,8- cineole were more abundant in wild plants exposed to a mechanical damage than cultivated plants. Hence, these compounds have been associated to induced defense, these results suggest that domestication reduced the induction of defensive volatiles in cultivated murtilla in response to mechanical damage.


Los compuestos volátiles están implicados en la defensa inducida contra insectos, desempeñando un papel importante en esta interacción. Sin embargo, estos compuestos podrían disminuir según el grado de domesticación. Actualmente, se ha reportado que algunos metabolitos secundarios son reducidos en plantas de murtilla domesticadas. Por lo tanto, surge la siguiente pregunta de investigación: ¿Los compuestos orgánicos volátiles inducidos por el daño mecánico son reducidos en plantas cultivadas de murtilla en comparación con plantas silvestres? Para dos ecotipos cultivados y sus respectivas contrapartes silvestres, los compuestos volátiles fueron capturados usando columnas de Porapak-Q y las muestras analizadas por cromatografía gaseosa. Los resultados mostraron que compuestos tales como 2- hexanona, α-pineno, 2-tujeno, 3-tujeno y 1,8-cineol fueron más abundantes en plantas silvestres expuestas a daño mecánico que en cultivadas. Debido a que estos compuestos se han asociado a defensa inducida, estos resultados sugieren que la domesticación reduce la inducción de volátiles en plantas cultivadas sometidas a daño mecánico.

ABSTRACT

Background: Cinnamomum longepaniculatum is an important commercial crop and the main source of volatile terpenoids. The biosynthesis of key bioactive metabolites of C. longepaniculatum is not well understood because of the lack of available genomic and transcriptomic information. To address this issue, we performed transcriptome sequencing of C. longepaniculatum leaves to identify factors involved in terpenoid metabolite biosynthesis. Results: Transcriptome sequencing of C. longepaniculatum leaves generated over 56 million raw reads. The transcriptome was assembled using the Trinity software and yielded 82,061 unigenes with an average length of 879.43 bp and N50 value of 1387 bp. Furthermore, Benchmarking Universal Single-Copy Orthologs analysis indicated that our assembly is 91% complete. The unigenes were used to query the nonredundant database depending on sequence similarity; 42,809 unigenes were homologous to known genes in different species, with an annotation rate of 42.87%. The transcript abundance and Gene Ontology analyses revealed that numerous unigenes were associated with metabolism, while others were annotated in functional categories including transcription, signal transduction, and secondary metabolism. The Kyoto Encyclopedia of Genes and Genomes pathway analysis showed that 19,260 unigenes were involved in 385 metabolic pathways, with 233 unigenes found to be involved in terpenoid metabolism. Moreover, 23,463 simple sequence repeats were identified using the microsatellite identification tool. Conclusion: This is the first detailed transcriptome analysis of C. longepaniculatum. The findings provide insights into the molecular basis of terpenoid biosynthesis and a reference for future studies on the genetics and breeding of C. longepaniculatum.

ABSTRACT Piper hispidinervum C. DC. is popularly known as long-pepper and it owns a commercial value due to the essential oil it produces. Long-pepper oil is rich in safrole and eugenoln components that have insecticidal, fungicidal and bactericidal activity. It has been establish that to medicinal plants light influences not only growth but also essential oil production. The growth, the content of photosynthetic pigments and the essential oil production of Piper hispidinervum at greenhouses with different light conditions was evaluated. The treatments were characterized by cultivation of plants for 180 days under different light conditions, produced by shading greenhouses with 50% and 30% of natural incident irradiance, two colored shading nets red (RN) and blue (BN) both blocking 50% of the incident radiation and one treatment at full-sun (0% of shade). The results showed that the treatments of 50% shade and RN and BN were the ones which stimulated the greater growth. Blue and red light also had the best production of photosynthetic pigments. Essential oil yielded more under full sun therefore this is the most indicated condition to produce seedlings for the chemical and pharmaceutical industry.

Os óleos essenciais (OEs) são metabólitos secundários que podem apresentar potencial no controle alternativo de nematoides. Assim objetivou-se testar o OE de pimenta rosa no controle de P. zeae em condições in vitro e na cultura do milho. O OE foi extraído por hidrodestilação, utilizando massa fresca das folhas e frutos maduros. O experimento in vitro foi instalado em arranjo fatorial 8x2, sendo oito concentrações (0,00, 7,80, 15,00, 31,00, 62,00, 125,00, 250,00 e 500,00 µg mL-1) e OE das folhas e dos frutos. As unidades experimentais foram tubos de ensaio com capacidade para 5mL, contendo 1 mL de cada concentração, e 1mL de suspensão de 50 nematoides. Decorrido 48 horas avaliou-se a porcentagem de mortalidade. O experimento em casa de vegetação foi instalado em arranjo fatorial 5x2, sendo cinco concentrações (0,00, 7,8, 15,0, 31,0 e 62,0 µg mL-1) e OEs (Folhas e frutos). As unidades experimentais constituíram-se por vasos com capacidade para 1,5 kg, os quais foram inoculados com 2000 e 1800 nematoides e semeados com sementes de milho tratadas com as concentrações dos OEs. Os OEs e as concentrações avaliadas apresentaram porcentagem de mortalidade em condições in vitro e reduziram a população de P. zeae, em casa de vegetação, o OE dos frutos promoveu maior mortalidade nos testes in vitro. Sendo assim os OEs demonstraram potencial no controle de fitonematoide, porém, há a necessidade de estudos a aplicabilidade dos mesmos.(AU)


Essential oils (EOs) are secondary metabolites that may present potential for an alternative control of nematodes. The purpose of this study was to test the EO of pink peppercorn in the control of P. zeae under in-vitro conditions and in the maize crop. Essential Oil was extracted by hydro-distillation using fresh leaf mass and mature fruits. The in-vitro experiment was installed in an 8x2 factorial arrangement, with eight concentrations (0.00, 7.8, 15.0, 31.0, 62.0, 125.0, 250.0 and 500.0 µg mL-1) and EO from leaves and fruits. The experimental units were 5-mL test tubes containing 1 mL of each concentration and 1 mL of suspension of 50 nematodes. The mortality percentage was evaluated after 48 hours. The greenhouse experiment was installed in a 5x2 factorial arrangement, with five concentrations (0.00, 7.8, 15.0, 31.0 and 62.0 µg mL-1) and EOs (leaves and fruits). The experimental units consisted of 1.5-kg capacity vases, which were inoculated with 2000 and 1800 nematodes, and seeded with corn seeds treated with EO concentrations. The EOs and concentrations evaluated presented a mortality percentage under the in-vitro conditions and reduced the P. zeae population in the greenhouse, while the EO from the fruits promoted higher mortality in the in-vitro tests. Therefore, EOs have shown potential in the control of phytonematodes, but further studies are required for their applicability.(AU)


Los aceites esenciales (OEs) son metabolitos secundarios que pueden presentar potencial en el control alternativo de nematodos. Así se objetivó probar el OE de pimienta rosa en el control de P. zeae en condiciones in vitro y en el cultivo del maíz. El OE fue extraído por hidrodestilación utilizando masa fresca de las hojas y frutos maduros. El experimento in vitro fue instalado en un arreglo factorial 8x2, siendo ocho concentraciones (0,00, 7,80, 15,00, 31,00, 62,00, 125,00, 250,00 y 500,00 µg mL-1) y OE de las hojas y de los frutos. Las unidades experimentales fueron tubos de ensayo con capacidad para 5 mL, conteniendo 1 mL de cada concentración, y 1 mL de suspensión de 50 nematodos. Después de 48 horas se evaluó el porcentaje de mortalidad. El experimento en casa de vegetación fue instalado en arreglo factorial 5x2, siendo cinco concentraciones (0,00, 7,8, 15,0, 31,0 y 62,0 µg mL-1) y OEs (Hojas y frutos). Las unidades experimentales se constituyeron por floreras con capacidad para 1,5 kg, los cuales fueron inoculados con 2000 y 1800 nematodos y sembrados con semillas de maíz tratadas con las concentraciones de los OEs. Los OEs y las concentraciones evaluadas presentaron un porcentaje de mortalidad en condiciones in vitro y redujeron la población de P. zeae, en casa de vegetación, el OE de los frutos promovió mayor mortalidad en las pruebas in vitro. Así los OE demostraron potencial en el control de fitonematodos, sin embargo, hay la necesidad de estudios a la aplicabilidad de los mismos.(AU)

Abstract This study was developed to evaluate the effect of seasonality on the yield and chemical composition of the essential oil (EO) of Hesperozygis ringens (Benth.) Epling, a native species from the Brazilian Pampa. Leaves were collected from four specimens of a single population in each of the four seasons for a year and were extracted in triplicate by hydro-distillation for 2 hours. The yield of EO (% w/w) was calculated on fresh weight basis (FWB), and the 16 oil samples were analyzed by gas chromatography coupled to mass spectrometry (GC-MS) and gas chromatography with flame ionization detector (GC-FID). Hierarchical Cluster Analysis (HCA) and Principal Component Analysis (PCA) were used as statistical tools to evaluate differences in chemical composition. The highest yields were obtained in autumn, spring and summer (2.32-4.38%), while the lowest yields were detected in winter, ranging from 1.15 to 1.91%. Oxygenated monoterpenoids were the predominant class of chemical constituents in the EO obtained in all seasons, showing the highest contents in autumn and summer, and pulegone was identified as a major compound, whose contents varied between 54.13 and 81.17%. The EO samples were divided into three chemical groups by HCA and PCA and were assigned to the same group, except for the three samples gathered in winter. The results showed a seasonal influence on the yield and chemical composition of the EO.


Resumo Este estudo foi desenvolvido a fim de avaliar o efeito da sazonalidade no rendimento e composição química do óleo essencial (OE) de Hesperozygis ringens (Benth.) Epling., uma espécie nativa do Pampa brasileiro. Folhas foram coletadas de quatro indivíduos de uma mesma população, em cada uma das quatro estações de um ano, e foram extraídas em triplicada por hidrodestilação durante 2 horas. O rendimento do OE (% m/m) foi calculado considerando a base fresca (BF) e as 16 amostras de óleo foram analisadas por cromatografia gasosa acoplada à espectrometria de massas (CG-EM) e cromatografia gasosa com detector de ionização de chamas (CG-DIC). Análise Hierárquica de Cluster (AHC) e Análise de Componentes Principais (ACP) foram utilizadas como ferramentas estatísticas para avaliar as diferenças na composição química. Os maiores rendimentos foram obtidos no outono, primavera e verão (2,32-4,38%), enquanto que os menores foram detectados no inverno, variando de 1,15 até 1,91%. Os monoterpenoides oxigenados foram a classe predominante dos constituintes do OE obtido em todas as estações, apresentando os maiores teores no outono e no verão, e a pulegona foi identificada como o constituinte majoritário, cujos teores variaram entre 54,13 e 81,17%. As amostras de OE foram divididas em três grupos químicos por AHC e ACP e foram classificadas no mesmo grupo, com exceção de três amostras coletadas no inverno. Os resultados demonstraram influência sazonal no rendimento e na composição química dos OE.

ABSTRACT

Devido à necessidade da substituição dos antimicrobianos como melhoradores de desempenho por aditivos alternativos, inúmeros produtos estão em estudo. Dentre esses estão os óleos essenciais que apresentam características antimicrobianas, contribuindo para manutenção da integridade intestinal e consequentemente melhora no desempenho, garantindo a segurança alimentar, evitando a resistência cruzada, bem como a contaminação do meio ambiente. Dessa forma, objetivou-se avaliar os efeitos da inclusão de uma mistura de óleos essenciais extraídos de orégano, alho, limão, alecrim, timo, eucalipto e laranja doce, na dosagem de 300 mL para 500 L de água de bebida, sobre a morfometria da mucosa do duodeno de frangos de corte. Foram avaliados quatro aviários tratados com óleos essenciais na ração e quatro aviários controle, durante o período de um a sete dias de idade, contendo pintainhos da linhagem Cobb. Para a análise morfométrica, cinco aves de cada aviário (total de 40 aves) foram mortas, utilizando-se de protocolo anestésico, medicação pré-anestésica de xilazina 4mg/Kg IM. e após solução de tiopental na dose de 25 mg/Kg IV. Após eutanásia, foram colhidas amostras de material do intestino delgado, correspondente ao segmento duodenal, para realização das medições de altura de vilo e profundidade de cripta. Não foram verificados efeitos sobre a altura de vilo e profundidade de cripta nos animais tratados com óleos essenciais (P>0,05). Nas condições em que foram realizados o experimento, pode-se concluir que os óleos essenciais não alteraram a altura de vilo e a profundidade de cripta do duodeno.


Due to the need for replacement of antimicrobials as performance enhancers to alternative additives, several products are being studied. Among them, essential oils that exhibit antimicrobial properties are being analyzed, which contribute to the maintenance of intestinal integrity and consequently improving performance, assuring food safety, preventing cross-resistance, as well as contamination of the environment. Thus, the objective of this study is to evaluate the effects of adding a mixture of essential oils from oregano, garlic, lemon, rosemary, thyme, eucalyptus and sweet orange, at a dosage of 300 mL for 500 L of drinking water, on the morphometry of duodenal mucosa of broilers. Four poultry aviaries treated with essential oils in the diet and four control poultry aviaries were evaluated during the period from one to seven day old, containing Cobb chicks. For the morphometric analysis, five birds in each aviary (total of 40 birds) were euthanized using anesthetic protocol premedication with xylazine 4mg/kg IM) and after, a solution of thiopental (25 mg /Kg IV). After euthanasia, material samples were collected from the small intestine (duodenum) for measuring villus height and crypt depth. No effects on villus height and crypt depth could be found in the animals treated with essential oils (P> 0:05). Under the experimental conditions, it can be concluded that essential oils did not alter villus height and crypt depth of the duodenum.


Debido a la necesidad de reemplazo de antimicrobianos como mejoradores de desempeño por aditivos alternativos, innúmeros productos están siendo estudiados. Entre esos están los aceites esenciales que presentan características antimicrobianas, lo que contribuye al mantenimiento de la integridad intestinal y por consiguiente mejora el rendimiento, por garantizar la seguridad alimentaria, evitando resistencia cruzada, así como la contaminación del medio ambiente. Por lo tanto, el objetivo ha sido evaluar los efectos de la adición de una mezcla de aceites esenciales extraídos del orégano, ajo, limón, romero, tomillo, eucalipto y naranja dulce, a una dosis de 300 ml para 500 litros de agua potable, sobre la morfometría de la mucosa duodenal de pollos de engorde. Se han evaluado cuatro aviarios tratados con aceites esenciales en el pienso y cuatro aviarios control, durante el período de uno a siete días de edad, conteniendo pollitos Cobb. Para el análisis morfométrico, cinco aves de cada aviario (total de 40 aves) fueron sacrificadas, utilizándose de protocolo anestésico, medicación pre anestésica de xilazina 4 mg /Kg IM., y después tiopental solución a una dosis de 25 mg /Kg IV. Después de la eutanasia, se ha recogido muestras de material del intestino delgado, que corresponde al segmento duodenal, para llevar a cabo las mediciones de la altura de las vellosidades y profundidad de la cripta. No se ha verificado efectos sobre la altura de las vellosidades y la profundidad de la cripta en los animales tratados con aceites esenciales (P> 0,05). En las condiciones en que se han realizado el experimento, se puede concluir que los aceites esenciales no alteraron la altura de las vellosidades y la profundidad de la cripta del duodeno.

ABSTRACT

Diversos cosméticos têm sido produzidos para atender a crescente demanda de mercado. Para que estes produtos possuam boa aceitação são necessárias que os mesmos apresentem adequadas características físicoquímicas e sensoriais, bem como eficácia, segurança e qualidade. Esse trabalho visou o desenvolvimento e avaliação da estabilidade de formulações biocosméticas a partir da incorporação dos óleos essenciais da espécie Lippia gracilis Schum em formulações cosméticas para cabelo, por meio de estudos de pré-formulação e testes de estabilidade preliminar e acelerada. Foram realizados testes físico-químicos para avaliação dos parâmetros: pH, viscosidade aparente, características sensoriais, espuma e densidade. Durante os estudos de estabilidade preliminar e acelerada, foi observado que as temperaturas: Refrigerador (TG) (5,0 ± 0,5ºC) e Estufa (TE) (45,0 ± 0,5ºC) sofreram alterações de pH, viscosidade e das características sensoriais. Ao final do estudo, mantiveram as características iniciais, apenas as formulações mantidas em temperatura ambiente. Concluiu-se que a formulação de xampu não possui estabilidade quando exposta a temperaturas abaixo de 5ºC e acima de 45ºC. A formulação de condicionador, além de ser instável em temperaturas extremas, apresentou também alterações após 30 dias (T2) em temperatura ambiente (TA) (24,0 ± 2,0ºC).(AU)


Several cosmetics have been produced to meet the growing market demand. For these products have good acceptance is necessary that they show suitable physico-chemical and organoleptic characteristics, as well as efficacy, safety and quality. This research aimed at the development and evaluation of the stability of formulations biocosmetics from the incorporation of the essential oils of the species Lippia gracilis Schum in shampoo and conditioner through preliminary stability testing and accelerated. Physico-chemical tests for evaluation of the parameters were performed: pH, viscosity, sensory characteristics, lather and density. During the stability studies and preliminary accelerated, it was observed that the formulations subjected to extreme temperatures, such as: Refrigerator (TG) (5.0 ± 0.5 ° C) and hot air oven (TE) (45.0 ± 0.5 ° C), undergone changes in pH, viscosity and sensory characteristics. At the end of the study maintained the initial characteristics, only formulations maintained at room temperature. It was concluded that the shampoo formulation lacks stability when exposed to temperatures below 5 ° C and above 45 ° C. And the conditioner formulation in addition to being unstable in extreme temperatures, also showed changes after 30 days (T2) at room temperature (RT) (24.0 ± 2.0 ° C).(AU)

ABSTRACT

Human respiratory syncytial virus (HRSV) is an important respiratory pathogens among children between zero-five years old. Host immunity and viral genetic variability are important factors that can make vaccine production difficult. In this work, differences between biological clones of HRSV were detected in clinical samples in the absence and presence of serum collected from children in the convalescent phase of the illness and from their biological mothers. Viral clones were selected by plaque assay in the absence and presence of serum and nucleotide sequences of the G2 and F2 genes of HRSV biological clones were compared. One non-synonymous mutation was found in the F gene (Ile5Asn) in one clone of an HRSV-B sample and one non-synonymous mutation was found in the G gene (Ser291Pro) in four clones of the same HRSV-B sample. Only one of these clones was obtained after treatment with the child's serum. In addition, some synonymous mutations were determined in two clones of the HRSV-A samples. In conclusion, it is possible that minor sequences could be selected by host antibodies contributing to the HRSV evolutionary process, hampering the development of an effective vaccine, since we verify the same codon alteration in absence and presence of human sera in individual clones of BR-85 sample.

ABSTRACT

RESUMOPatchouli é uma espécie aromática pertencente à família Lamiaceae que possui óleo essencial com alto valor comercial devido à aplicação nas indústrias de higiene, cosméticos e perfumarias. Considerando as características voláteis e estruturas de armazenamento em diferentes órgãos das plantas aromáticas, a determinação de métodos de secagem é de extrema importância e pode afetar tanto o rendimento como a composição do óleo essencial. Este trabalho teve como objetivo principal avaliar o teor e a composição do óleo essencial de patchouli após secagem por diferentes períodos em estufa e a temperatura ambiente. O experimento foi realizado durante o mês de fevereiro de 2011 em delineamento experimental inteiramente casualizado em esquema fatorial 2X6, correspondente a dois métodos de secagem (estufa 65ºC e temperatura ambiente) e seis tempos para o início da extração do óleo essencial após a (0, 2, 4,6,8 e 10 dias após a colheita),com três repetições. O óleo essencial foi extraído por meio hidrodestilação em aparelho graduado de Clevenger e os constituintes do óleo essencial foram analisados por cromatografia em fase gasosa acoplada à espectrometria de massas. Houve interação significativa entre os métodos e tempos de secagem tanto para o teor como para a composição do óleo essencial. A secagem das plantas por dois dias em estufa a 65ºC aumentou o teor de óleo essencial, após este período houve redução significativa. Para as folhas secas em temperatura ambiente, o teor de óleo essencial foi superior aos quatro e aos seis dias após a colheita. Comparando-se os métodos, a secagem em estufa mostrou-se mais eficiente, sendo o teor significativamente superior após dois dias de secagem. Após este período, no entanto, houve drástica redução do teor de óleo essencial, sendo inferior ao teor observado em todos os períodos nas folhas secas em temperatura ambiente. Os diferentes métodos e tempos de secagem alteraram também a composição do óleo essencial. Embora a secagem em estufa a 65º por dois dias não tenha resultado em redução dos teores dos constituintes majoritários patchoulol e pogostol, maiores teores foram observados após seis ou oito dias de secagem. Quando a secagem foi realizada a temperatura ambiente, não houve alteração dos teores de ambos os constituintes.


ABSTRACTPatchouli is an aromatic species belonging to the Lamiaceae family that has essential oil with high commercial value to the hygiene, cosmetic and perfume industries. Considering its volatile characteristics and storage structures in different organs of aromatic plants, the determination of drying methods is extremely important and can affect both the yield and the composition of the essential oil. This work aimed to evaluate the content and composition of the patchouli essential oil after drying it in different periodin an oven and at room temperature. The trial was carried out in February 2011 at an experimental design completely randomized in 2X6 factorial scheme, corresponding to two drying methods (oven at 65˚C and room temperature) and six periods of essential oil extraction after harvest (0, 2, 4, 6, 8, and 10 days after harvest), with three replications. The essential oil was extracted by hydrodistillation in a Clevenger apparatus and the essential oil components were analyzed by gas chromatography coupled to mass spectrometry. There was significant interaction among the methods and drying times for both the content as well as for the composition of essential oil. The drying of the plants for two days in an oven at 65˚C increased the essential oil content; after this period, there was a significant reduction. For the leaves that dried at room temperature, their contentof essential oil was superior at the fourth and sixth days after the harvest. When comparing the methods, the drying in the oven was more efficient, with the oil content being significantly higher after two days of drying. After this period, however, there was a drastic reduction in the essential oil content, lower than the one observed in all periods when the leaves dried at room temperature. The different methods and drying periods also altered the essential oil`s composition. Although drying in oven at 65˚ for two days did not result in reduction of the contents of patchoulol and pogostol, higher contents were observed after six or eight days of drying. When drying was carried out at room temperature, no change in relative content of both components was observed.

This study aims at evaluating the effects of Zataria multiflora (Z. multiflora) essential oil (EO) on growth, aflatoxin production and transcription of aflatoxin biosynthesis pathway genes. Total RNAs of Aspergillus parasiticus (A.parasiticus) ATCC56775 grown in yeast extract sucrose (YES) broth medium treated with Z. multiflora EO were subjected to reverse transcription-polymerase chain reaction (RT-PCR). Specific primers of nor-1, ver-1, omt-A and aflR genes were used. In parallel mycelial dry weight of samples were measured and all the media were assayed by high-pressure liquid chromatography (HPLC) for aflatoxinB1 (AFB1), aflatoxinB2 (AFB2), aflatoxinG1 (AFG1), aflatoxinG2 (AFG2) and aflatoxin total (AFTotal) production. The results showed that mycelial dry weight and aflatoxin production reduce in the presence of Z. multiflora EO (100 ppm) on day 5 of growth. It was found that the expression of nor-1, ver-1, omt-A and aflR genes was correlated with the ability of fungus to produce aflatoxins on day 5 in YES medium. RT-PCR showed that in the presence of Z.multiflora EO (100 ppm) nor-1, ver-1 and omtA genes expression was reduced. It seems that toxin production inhibitory effects of Z. multiflora EO on day 5 may be at the transcription level and this herb may cause reduction in aflatoxin biosynthesis pathway genes activity.

ABSTRACT

The essential oil of the aerial parts of Senecio selloi Spreng. DC. was extracted by hydrodistillation and analyzed by GC/MS. Nineteen compounds were identified, representing 99.9% of the total. The main compounds were found to be sesquiterpene hydrocarbons (71.3%), most of them with a bisabolane skeleton (59.4%). The major constituent was α-zingiberene (54%), followed by monoterpene α-isolimonene (16%). The essential oil was also tested against two Gram-positive and two Gram-negative bacterial species, three yeasts, and an algae. From the strains assayed, only Bacillus subtilis ATCC 6633 showed susceptibility (MIC and MBC = 4400 µg/mL) to the essential oil.


O óleo essencial das partes aéreas de Senecio selloi Spreng DC. foi extraído por hidrodestilação e analisado por CG/EM. Dezenove constituintes foram identificados, representando 99,9% do total. Os principais compostos fornecidos foram sesquiterpenos hidrocarbonetos (71,3%), a maioria destes com esqueleto bisabolano (59,4%). O constituinte majoritário foi a-zingibereno (54%), seguido do monoterpeno a-isolimoneno (16%). O óleo essencial foi testado contra duas cepas Gram-positivas e duas Gram-negativas, três fungos e uma alga. De todas as linhagens testadas somente Bacillus subtilis ATCC 6633 mostrou suscetibilidade (CIM e CBM = 4400 µg/mL) para o óleo essencial.

ABSTRACT

Objetivou-se avaliar a produção de fitomassa, teor e produtividade do óleo essencial de Baccharis dracuculifolia DC., em função de doses de adubo orgânico, e foram utilizadas mudas produzidas por semente, e mantidas sob sombrite pelo período de 84 dias até serem transplantadas para o campo, no espaçamento de 1,0 x 1,0 m, com irrigação por gotejamento. O delineamento estatístico utilizado foi em blocos ao acaso, com quatro repetições, e as doses de composto orgânico foram: 0, 10, 20, 30, 40 e 50 t ha-1. Foi realizada uma colheita, aos 150 dias após o transplante das mudas. O óleo essencial foi extraído por hidrodestilação e analisado em cromatógrafo a gás acoplado a espectrômetro de massas (Shimadzu, QP-5000). Todos os resultados foram submetidos à análise de variância pelo teste F seguido da análise de regressão e o teste Tukey para os dados: massa seca útil da parte aérea, teor e produtividade do óleo essencial. Para as variáveis massa seca total e massa seca útil da parte aérea, as dosagens estudadas influenciaram as plantas, que apresentaram os melhores resultados na dose 50 t ha-1. Para teor de óleo não houve influência significativa para as dosagens estudadas, porém a dose 30 t ha-1 apresentou o melhor resultado. Entretanto para a produtividade do óleo essencial, as dosagens estudadas influenciaram as plantas, que na dosagem 30 t ha-1 obtiveram os melhores resultados.


The objective was to evaluate the characteristics of the Baccharis dracunculifolia DC., depending on levels of organic fertilizer, and seedlings were used B. dracunculifolia produced by seed, and kept under shade for a period of 84 days before being transplanted to the field, spaced 1.0 x 1.0 m, with drip irrigation. The experimental design was randomized blocks with four replications, and doses of organic compost, 0, 10, 20, 30, 40 and 50 t ha-1. We performed a harvest, 150 days after transplanting. The essential oil was extracted by hydrodistillation and analyzed by gas chromatography coupled to mass spectrometer (Shimadzu, QP-5000). All results were submitted to analysis of variance F test followed by regression analysis and Tukey test for the data: useful dry mass of shoots, content of essential and productivity of essential oil. For the variables total dry mass and dry mass of shoots useful, the doses studied influenced the plants, which showed the best results at a dose 50 t ha-1. For content of essential oil no significant influence was verified on the doses studied, but the dose 30 t ha -1 showed the best result. However the productivity of the essential oil, the doses studied influenced the plants, which in dose 30 t ha-1 obtained the best results.

ABSTRACT

Background & objectives: Development of insect resistance to synthetic pesticides, high operational cost and environmental pollution have created the need for developing alternative approaches to control vector-borne diseases. In the present study we have investigated the insecticidal activity of essential oil isolated from the leaves of Lantana camara against mosquito vectors. Methods: Essential oil was isolated from the leaves of L. camara using hydro-distillation method. Bioassay test was carried out by WHO method for determination of adulticidal activity against mosquitoes. Different compounds were identified by gas chromatography-mass spectrometry analysis. Results: LD50 values of the oil were 0.06, 0.05, 0.05, 0.05 and 0.06 mg/cm2 while LD90 values were 0.10, 0.10, 0.09, 0.09 and 0.10 mg/cm2 against Ae. aegypti, Cx. quinquefasciatus, An. culicifacies, An. fluvialitis and An. stephensi respectively. KDT50 of the oil were 20, 18, 15, 12, and 14 min and KDT90 values were 35, 28 25, 18, 23 min against Ae. aegypti, Cx. quinquefasciatus, An. culicifacies, An. fluviatilis and An. stephensi, respectively on 0.208 mg/cm2 impregnated paper. Studies on persistence of essential oil of L. camara on impregnated paper revealed that it has more adulticidal activity for longer period at low storage temperature. Gas chromatographic-mass spectrometric analysis of essential oil showed 45 peaks. Caryophyllene (16.37%), eucalyptol (10.75%), α-humelene (8.22%) and germacrene (7.41%) were present in major amounts and contributed 42.75 per cent of the total constituents. Interpretation &conclusion: Essential oil from the leaves of L. camara possesses adulticidal activity against different mosquito species that could be utilized for development of oil-based insecticide as supplementary to synthetic insecticides.
